Our basic course 2 „Basics in Optical Sectioning Microscopy (confocal, Deconvolution, TIRF, lightsheet)” is intended for all interested people who are already familiar with the basics of light microscopy which were covered in the Basics in Light Microscopy course (including the basic concepts of light, the light path in a transmission and reflection microscope, fluorescence and terms like resolution, magnification, reflection …), but would like to understand the concepts of optical sectioning, with a focus on point scanning confocal microscopy.
The lectures of the course will cover the following topic:

  • Basic principal of confocality
  • Different types of confocal microscopes (point scanner, slit scanner, and spinning disc, two photon)
  • Different components of a confocal microscopes (lasers, detectors, acousto-optical elements, prisms …)
  • Optical sectioning
  • Confocal imaging parameters (pixel dwell; time signal to noise; pin hole size …)
  • Basic principal of Single Plane Illumination (SPIM) (different types of light sheets, geometry of SPIM systems, research example)
  • Ideal conditions for live cell imaging (environmental control, media, sample mounting, autofocus strategies, phototoxicity, imaging conditions)
  • Basic principal of Total Internal Reflection Fluorescence (TIRF)

During the practical’s we will apply the basic concepts learned during the lectures.
